Tissue Culture Basics

This guide introduces the core elements of plant tissue culture, including culture media, environmental conditions, and common additives. Use it as a planning reference alongside your own protocols, plant material, laboratory setup, and trusted published sources.

Culture Media

Culture media provide water, mineral nutrients, vitamins, a carbon source, and other components that support plant material in vitro. Many tissue culture workflows begin with an established basal medium such as Murashige and Skoog (MS), Woody Plant Medium (WPM), or a laboratory-specific formulation.

Record formulation details such as components, concentrations, target pH, and preparation notes in the Media Recipe. Link the recipe to the relevant Plant or Explant, and record media on Transfers when applicable. Keep vessel, batch, Stage, and observation context with the culture record so Transfers, contamination records, and culture observations can be reviewed together over time.

  • Use water quality appropriate for your laboratory protocol.
  • Record the carbon source and concentration used in the formulation.
  • Record the target pH specified by the protocol before sterilization or other preparation steps.
  • Prepare and close culture vessels according to your established laboratory procedure.

Media Components to Track

A Media Recipe is easier to review and compare when its key formulation details are recorded consistently.

Base formulationStart with an established basal medium such as MS, WPM, or a laboratory-specific formulation, then document any changes made for the species, stage, or protocol.
Carbon sourceSucrose is a common carbon source in plant tissue culture. Record the carbon source and concentration used in each formulation.
Gelling and textureGelling agents such as agar or gellan gum can be used to create a semi-solid culture medium. Record the type and concentration because firmness and clarity can affect handling and observation.
pH and sterilizationRecord the target pH and preparation method used for each batch so differences can be reviewed alongside later culture outcomes.

Culture Conditions

Culture conditions vary by species, stage, and protocol. Record the environmental conditions used so culture responses can be compared over time.

  • Temperature range
  • Light level and photoperiod
  • Culture location or room
  • Any other environmental conditions relevant to the protocol

Additives and Experiments

Additives can influence culture development, rooting, tissue appearance, and other outcomes. Record them as formulation variables so changes can be reviewed against later observations.

Plant Growth RegulatorsPlant growth regulator requirements vary by species, tissue type, and stage. Record the compounds and concentrations used so responses can be compared across cultures.
Organic additivesAdditives such as coconut water, banana-derived materials, or plant extracts can vary by source and batch. Record the material, concentration, and source whenever they are used.
Activated charcoalActivated charcoal is used in some tissue culture formulations and can interact with multiple compounds in the medium. Record when it is used, its concentration, and the intended purpose.
Contamination observationsKeep contamination records separate from Media Recipe changes. This makes it easier to review whether contamination patterns are associated with preparation, handling, environment, or other workflow factors.
