Organizing Your First Tissue Culture
Learn how to organize a source plant, create explants, record stages, and build a connected tissue culture history from day one.
Start with a Plant Record
In xPlant, use a Plant record to represent the source plant for a tissue culture line. Preserving that lineage makes later comparisons — which batch grew well, where contamination was repeatedly observed, or how related explants developed — easier to interpret instead of leaving a string of disconnected notes. Record the source plant's identity: a name, its species, and a photo if you have one. The Plant record serves as the lasting source record for related Explants and remains in your lab history even after those Explants have been discarded. Create one from Dashboard → Plants → New Plant.
Tip: Use a consistent naming convention from the start, such as species + cultivar + batch or date. It makes finding and organizing plants much easier as your collection grows.
Establish an Explant
An Explant represents an individual tissue culture established from a source plant. Each Explant needs its own identity because it develops its own culture history while staying linked back to the source Plant. What matters most here is a descriptive label — Shoot Tip 1, Node A — that will still make sense months later when comparing outcomes across related Explants. In xPlant, add one from the Plant detail page via Add Explant.
Tip: Create one Explant for each culture you want to track independently. This keeps each culture's history distinct while preserving its link to the source Plant.
Record Initiation
For a new Explant, xPlant defaults the first Stage to Explant Initiation — the point where excised tissue is introduced to culture media and early establishment is observed. Stage names and sequences can vary by protocol, and Surface Sterilization is also available when your workflow records it as a separate Stage. Record the date, any observations, and the room or bench location if your lab tracks physical space. Early observations provide useful context when reviewing how the culture develops later.
Tip: Record observations at every stage, even if they're brief. A note like "Healthy initiation, no browning" becomes valuable reference material later.
Record the Next Stage
In xPlant, culture development can be recorded through named Stages such as Explant Initiation, Multiplication, Rooting, and Acclimatization. Stage names and sequences can vary by protocol. When a culture enters a new Stage, create a new Stage record rather than replacing the previous one. Capture the Stage status, dates, observations, and room context. Link relevant media recipes and SOPs to the Explant, and record media on Transfers when applicable, so that context can be reviewed alongside the Stage history. Add one from the Explant detail page via Add Stage.
Tip: Reference a media recipe and SOP when they apply. Together, they keep formulation and procedure context connected to the culture history.
Record a Transfer
When culture material is split, moved to another vessel, or subcultured, you can record that movement as a Transfer to keep the propagation history traceable alongside Stage changes. Record where the material came from, where it went, and when the Transfer occurred. In xPlant, Transfers are logged separately from Stages so movement and subculture history can be reviewed alongside culture development. Record one from the Explant detail page or the dashboard's Transfers section.
Tip: Transfers are optional. If your lab only tracks stage progression, simply adding new stages may be all you need.
Review the Culture History
Once a few Stages and Transfers exist, the value of recording them becomes easier to see: comparing outcomes associated with different protocols, reviewing whether contamination observations recur across a line, or comparing related Explants that took different paths. xPlant keeps previous Stages, observations, and Transfers connected to the Explant as its current state changes. The dashboard surfaces active cultures, current stages, and contamination flags, with search and filters to find a Plant or Explant by name, species, stage, or label.
Tip: Review culture histories regularly to compare outcomes, revisit protocol context, and use your records to inform future tissue culture work.
Recordkeeping principles
- Start simple: a complete record beats a perfect one. Begin with the plant, the explant, and its first stage, then layer in media, SOPs, and comments as the workflow becomes familiar.
- Record contamination as data, not a failure to hide. Log it as soon as you notice it, on the plant or explant it affected, so patterns across batches and media recipes become visible over time.
- Build records over time: a culture record becomes more useful as its history grows. A single Stage provides limited context; a longer history across Stages, Transfers, media, and observations gives you more context for reviewing what happened and comparing future work.
- Keep recordkeeping consistent: the same labeling and observation habits across every explant are what make comparisons possible later. Pick a convention early and keep using it, even for cultures that seem too simple to bother.
